Does ipamorelin raise cortisol?
There is a real paper behind the claim, from 1998, and this site cites it — it is not a marketing invention. What the paper says is narrower than the sentence it has turned into. It was measured in pigs, the cortisol half of it names something it was measured against, and the same question has no answer here for the other 4 molecules on this site at all.
Where the claim comes from
Raun et al. 1998 is the origin, and its title is a claim in itself: “Ipamorelin, the first selective growth hormone secretagogue”. The selectivity work is one part of a longer pharmacology paper, and the paper says in one sentence where that part was done.
The specificity for GH release was studied in swine.
Swine, then — conscious swine, as the study describes them elsewhere. That is not a disqualification and this page is not making it one: a pig pituitary is a reasonable place to ask this question, and the later writing this site has read repeats the finding rather than overturning it. It is a qualifier, and the value in this site’s own table carries it in the cell rather than in a footnote, which is why the row reads the way it does in the margin here.
What made the result worth reporting is what the comparators did in the same experiment.
Administration of both GHRP-6 and GHRP-2 resulted in increased plasma levels of ACTH and cortisol.
Two other growth hormone secretagogues put ACTH and cortisol up. The finding is that this one did not do what those did — a contrast, drawn inside one experiment, which is a much better piece of evidence than an isolated null result and also a much narrower claim than “does not raise cortisol” on its own.
The two halves of it are not equally strong
The recorded value names cortisol and prolactin together. The paper states them in two separate sentences, and the sentences are not the same strength. The prolactin one is flat and covers every compound tested.
None of the GH secretagogues tested affected FSH, LH, PRL or TSH plasma levels.
Nothing moved, in any of them. The cortisol sentence is built differently, and the difference is its closing clause.
Very surprisingly, ipamorelin did not release ACTH or cortisol in levels significantly different from those observed following GHRH stimulation.
“Not significantly different from those observed following GHRH stimulation” is a comparison, and what it compares to is another stimulus rather than an untreated animal. Read strictly, the sentence says this molecule did not raise ACTH or cortisol more than GHRH did. Whether GHRH itself moved them is a question this sentence does not answer, and this site has not read a document that does.
The margin the result held over is stated in the next sentence of the same abstract, and it is a wide one.
This lack of effect on ACTH and cortisol plasma levels was evident even at doses more than 200-fold higher than the ED50 for GH release.
What the human literature is quoted as saying instead
This site’s cell carries a second document beside the first, and it is there because the two say different things. A 2026 review, Dominikowski et al. 2026, characterises the same property in people as “comparatively minimal activation of other pituitary–adrenal axes in available human studies”.
“Comparatively minimal” and “did not release” are not the same claim, and picking the friendlier of them would be resolving a difference the reader is entitled to see. The swine result is where the claim comes from and it stays; the human characterisation is weaker and is printed beside it rather than instead of it. That is the same rule this site applies to three FDA labels that give three different half-lives: where two sources disagree, print both.
And on the other 4 molecules this site records nothing at all
Here is the part that says most about how far this site can answer the question at all. 1 of the 5 molecules recorded here carries anything on cortisol and prolactin. The other 4 — Sermorelin, Tesamorelin, CJC-1295 (no DAC), CJC-1295 + DAC — read “not recorded”, and that cell takes no citation marker, because no outside reference would settle it. What would settle it is us doing the work.
An unfilled cell is an open question and it is printed as one rather than left blank for a reader to fill in with an assumption. In particular it does not mean the other 4 raise cortisol and it does not mean they do not. It means this site has read no document either way, which is a smaller and more useful thing to say.
